High levels of phenylacetate, the active metabolite of phenylbutyric acid, can cause nausea, headache, emesis, fatigue, weakness, lethargy, somnolence, dizziness, slurred speech, memory loss, confusion, and disorientation.L42105

In the event of an overdose, discontinue the drug and institute supportive measures. Phenylbutyric acid is a fatty acid and a derivative of butyric acid naturally produced by colonic bacteria fermentation. It demonstrates a number of cellular and biological effects, such as relieving inflammation and acting as a chemical chaperone.A249035 It is used to treat genetic metabolic syndromes, neuropathies, and urea cycle disorders.L386,L42105

Metabolisme

The major sites for metabolism of sodium phenylbutyrate are the liver and kidney.L386 Phenylbutyric acid is rapidly metabolized to phenylacetate via beta-oxidation. Phenylacetate is conjugated with phenylacetyl-CoA, which in turn combines with glutamine via acetylation to form phenylacetylglutamine.L42115

Rute Eliminasi

Approximately 80–100% of the dose was excreted by the kidneys within 24 hours as the conjugation product, phenylacetylglutamine. For each gram of sodium phenylbutyrate administered, it is estimated that between 0.12–0.15 grams of phenylacetylglutamine nitrogen are produced.L386
